Original Comments The genomic ken[+] transgene was constructed by inserting the genomic sequence between nt. 8959(HindIII) and nt. 18,011(PstI) into the pCasper3 vector (the nucleotide numbers were defined in the genomic map described in Lukacsovich et al., 1999, Genomics 57, 43-56). The genomic ken[+] completely rescued the ken mutant phenotypes, unlike the hs-ken[+] and UAS-ken[+] transgenes. The gemomic region surrounding the ken locus is extremely dence in genes; 10 genes have been identified molecularly within the 45 kB stretch. Some of them physically overlap with each other. Thus the genomic ken[+] line would be useful to distinguish these adjacent transcription units by means of genetic complementation tests. The genomic ken[+] transgene inserted into X (DGRC# 109043, original# kN14-C1), 2nd (DGRC# 109042, original# kN07-D4) and third (DGRC# 109044, original# kN14-D1) chromosomes was equally effective in rescuing the ken mutant phenotypes. The stock with the 2nd chromosome insertion of the genomic ken[+] transgene also carries the P{BmDelta-w}ken[ok] mutation on the same chromosome.
