Young genes

Young genes with mutant or insertion stocks
From analysis of the 12 Drosophila species genomes,
Zhang et al. (2010) identified 947 young
protein-coding genes that were created recently in the
lineage leading to D. melanogaster. We listed
D. melanogaster stocks that are assigned to 412
"very young" genes.

GAL4 enhancer trap lines
NP lines (GETDB lines) are GAL4 enhancer trap lines (P{GawB} element-insertion lines) of
which 3' flanking sequences of the inserted element,
expression pattern and affected gene(s) were examined. We
have about 4,300 lines available for distribution.

GAL4 / GAL80
|
|
GAL4: Stage- and/or tissue-specific, or non-specific expression
GAL4 insertion line expresses the yeast GAL4
transcriptional activator under the control of a native
gene promoter, or driver gene in non-specific or
stage-and/or tissue-specific patterns. This collection
consists of GAL4 insertion and GAL4 enhancer trap lines
useful for UAS-mediated expression of a target gene.

GAL80 blocks GAL4 activity
The GAL80 blocks the GAL4's transcription activation
ability. It is useful to regulate expression using GAL4-UAS
system.

GS lines
|
|
Gene Search System: Forced expression induced by GAL4
GS lines are insertion lines of the P{GS} element (gene
search vector) and generated by the Drosophila Gene Search
Project organized by Toshiro Aigaki, Department of
Biological Sciences, Tokyo Metropolitan University. In the
vectors, there are some variation according to the number
and direction of UAS, availability of GFP marker or
Cre-loxP flipping system (
P{GSV1}
,
P{GSV2}
,
P{GSV3}
,
P{GSV6}
and
P{GSV7}
). All versions have ability to transcribe adjacent
genomic region forcedly depending on the GAL4
expression. All insertions were mapped to the genome
sequence, which can be searched by gene symbol, CG
number, CT number, or cytological map position. We have
about 6,900 lines available for distribution.

LA lines
|
|
Mae-UAS.6.11 Insertions: Misexpression induced by GAL4
Collection of
P{Mae-UAS.6.11} insertion lines. P{Mae-UAS.6.11}
carries y[+] marker and is designed to be used for
modular misexpression screen. GAL4-activated
transcription from Hsp70 promoter is toward P5',
extending into adjacent genomic
sequences.

UAS
|
|
UAS responders
The strains carrying a construct including upstream
activating sequence (UAS, β-gal-coding region
downstream from the GAL4-binding sites). A responder
gene downstream of UAS is expressed by crossing with a
strain expressing GAL4, driven by specific promoters or by
enhancer traps.
Some balancer chromosomes also have a UAS construct but
they are not in this subset. Strains carrying a
misexpression vector that includes UAS (GS lines and LA
lines) are NOT in this subset.

|
|
GFP fusion protein expression lines (Protein Trap)
Created by a GFP exon containing vector (Morin et al.,
2001). DNA sequence and subsequent mapping information from
the inserted P-element, and expression patterns of the
tagged proteins were examined. We have about 360 lines
available for distribution.

GFP, YFP, RFP etc.

Fluorescent protein insertion lines
GFP, YFP, DsRed and others insertion lines are
available. This collection is useful for studying protein
trafficking and localization. One of the main lines in this
collection, which has the inserted construct consisting of
either the UAS fused with GFP/YFP/DsRed alone, or the UAS
fused with GFP/YFP/DsRed and the coding region of a gene of
interest. The expression of UAS-reporter gene construct is
driven by the GAL4 driver. Also available are lines which
have the inserted construct of a gene's natural or
inducible promoter fused with GFP/YFP/DsRed. More than 100
lines are available for distribution.

|
|
phiC31 Integration System Components
The "starter kit" lines for transformation by site-specific
integration via phiC31-mediated recombinase-mediated
cassette exchange. Recombination between attachment sites
(an attP in fly genome of the stocks and an attB carried by
an integration vector) is catalyzed by the phage phiC31
integrase (

).

P element mutagenesis
|
|
P element mutagenesis starter kits and controls
Strains to start mutagenesis with P element. This
collection includes strains with Δ 2-3 (Delta2-3,
P{ Δ2-3}99B
) that allows expression of P transposase in somatic as
well as germ cells, and M, P and Q strains collected from
natural populations.

Mapping

Marker strains useful for recombination (classical) mapping
The series of strains are very useful for recombination
(classical) mapping. The strains carry visible mutations as
markers, for a specific chromosome and multiple
chromosomes. About 100 lines are available for
distribution.
This is NOT a kit for mapping. Proper choice is recommended
for your study .

Balancers

|
Stocks carrying balancer(s)
Balancers are chromosomes with inversions and dominant
marker mutations useful to maintain a lethal or sterile
homologous chromosome or a segment thereof intact in a
cross or stock. Balancer chromosomes are also useful for
the substitution of chromosomes between stocks.
The list will help the users make a choice what stocks to
get.

MARCM
|
|
Mosaic Analysis with a Repressible Cell Marker system
This collection consists of about 2,200 strains possessing
the autosomal insertions of a new piggyBac vector, made by
the laboratory of Professor L. Luo at Stanford University.
The strains have following properties:
- All strains have a insertion inside independent
transcriptional units.
- All insertions are on FRT-bearing chromosomes.
- The piggyBac vector have splicing acceptor and a
termination of translation in all three frames, thus
improving mutagenicity.
- The marker on the vector is a DsRed protein, enabling
easier viewing of mosaic clones labeled with GFP.

FRT-lethal (BruinFly)

|
Lethal lines with FRT and ey-FLP
Each stock has a lethal P-element insertion and a FRT
element on the 2L, 2R, 3L or 3R chromosome arm. Somatic
recombinations by using FLP generates mosaics of homozygous
for a lethal P-element, heterozygous and homozygous for
wild type. We have about 1,300 lines available for
distribution.

|
FRT (FLP recombination target) strains
These lines contain FRT sites that recognized by FLP
recombinase (see FLP). The
FLP-recombinase (Flippase) catalyzes recombination between
FRTs resulting in somatic clones.

|
Flipase producing lines
These lines contain a construct expressing the yeast
FLP-recombinase (Flippase) under the control of a gene
enhancer or promoter. The Flippase catalyzes recombination
between FRTs resulting in somatic clones.

DrosDel lines
|

Deletion lines generated by FRT-mediated recombination
This is a collection of molecularly mapped deletions of
Drosophila melanogaster on an isogenic background,
covering 77% of the genome, generated using FLP-mediated
recombination between pairs of FRT sites. About 500 lines
are available.

RS lines (RS3 and RS5)
|
|
FRT-carrying insertions can be used as docking sites
RS3 and RS5 FRT-carrying insertions can be used as docking
sites for FLP-mediated transgene remobilisation, using the
split white+ marker strategy. The
DrosDel deletions were generated by using RS lines. The
stocks are from the Szeged Drosophil Stock Centre, Szeged
University, Hungary.

DrosDel Core Deletion Kit: modified by DGRC Kyoto
Each kit is a set of deficiency lines for each chromosome
defined by the DrosDel project.
Since some stocks are not kept in DGRC Kyoto, we offer a
modified DrosDel Core Deletion Kit. The total coverage
becomes smaller than the original.
All deficiencies are molecularly mapped.

Deficiencies covers about 90% of the genome, provide
maximal coverage of the genome with the minimal number of
