| Original Comments | Promoter region of DmelObp57d (641bp; Yasukawa et al., 2010 Gene 467: 25-34. doi: 10.1016/j.gene.2010.07.006) was PCR amplified with primers 5'-AAAGAATTC-AGCCACAAACTGGAGGACAG-3' and 5'-AAAGGATCC-CAAACTAGTTGAAGATATCATAGGAAACT-3'. After digestion with EcoRI and BamHI, the promoter fragment was ligated into pCaSpeR3 with a BamHI-NotI Gal4 fragment from pGaTB (Phelps & Brand, 1998 METHODS: A Companion to Methods in Enzymology 14: 367-379. doi: 10.1006/meth.1998.0592). Introduced directly into w[1118]; Obp57d+e[KO]. Expression is observed in the cells associated with the Gr32a expressing neurons on 5th and 4th tarsal segment on each leg (Koganezawa et al., 2010 Current Biology 20: 1-8. doi: 10.1016/j.cub.2009.11.038). DmelObp57d>Gal4 #2 (P{w[+mC]=Obp57d-Gal4.M}TM2, DGRC# 109683) has broader expression (See also DmelObp57d>Gal4 #1 (P{w[+mC]=Obp57d-Gal4.M}TM1, DGRC# 109682). In addition to Obp57d expressing cells, it has expression in the cells associated with taste neurons on labella and wings. |
